Tocompletetheassay,standardorsamplecontainingnephrininthesolublephaseisaddedtomicrotiterwellsthatcontainnephrinantigenimmobilizedontothesolidphase,andtowhichrabbitanti-nephrinantibodyisaddedinthefluidphase.Theantibodybindstoeithertheimmobilizednephrinorthesolublenephrin,whichcompeteforthisbinding.Afteraonehourincubationatroomtemperature,theplatesarewashedandagoatanti-rabbit–HRPconjugateisusedtodetectboundantibody.Platesarewashedandonlytheantibodyconjugateboundtothestationaryphaseantigenremainsinthewell,whichisdetectedwithachromogenicreaction.Colorintensityisinverselyproportionaltotheconcentrationofnephrininthefluidphase.Concentrationsinexperimentalsamplesaredeterminedfromastandardcurve. |
