TheassayisbasedupontheJaffe"reactionofalkalinepicratewithcreatinine.Standardsorsamplesareaddedtoamicroplate,andalkalinepicratereagentisadded.Absorbanceat500nmisdeterminedaftera10minuteincubationonthebenchtop.Subsequently,anacidsolutionisadded,andabsorbanceisagaindeterminedaftera5minuteincubation.Thedifferencebetweentheseabsorbancevaluesisdirectlyproportionaltothethecreatinineconcentration,andastandardcurveisgeneratedfromtheresponsetothestandards.Unknownsamplesareevaluatedbycomparingresponsetothestandardcurve.Theassaymaybecompletedinlessthan30minutes.

Theassayservesasanormalizationprocedureforotheranalytesinurinesamples,particularlyforurinaryalbumin(1,2).Itallowsforameasureofkidneyfunctionbasedonaspoturinesampleratherthantheusual24hourpool.
