Summaryofprocedure:TheFibronectinELISAisasandwichassayforthequantitativemeasurementoffibronectininfreshlydrawnhumanplasma.Theassaydependsuponthebindingoffibronectin,instandardorinplasma,tospeciallycoatedmicrotiterwells.Theplateiswashedtoremoveunboundreactants,andfibronectinisdetectedwithananti-fibronectinantibody-HRPconjugate.Theplateisagainwashed,andTMBisaddedtodetectboundHRP.Thechromogenicreactionisstoppedwithdilutesulfuricacid,andcolorintensitydeterminedusingamicroplatereaderstetat450nm.Absorbanceisdirectlyproportionaltothelogarithmoffibronectinconcentrationaddedinthesample.Theassaymaybecompletedinlessthan3hours.
